5-Amino-1MQ (5-amino-1-methylquinolinium) is a synthetic small-molecule inhibitor of the NNMT (nicotinamide N-methyltransferase) enzyme. By blocking NNMT, it aims to prevent the breakdown of NAD+ and elevate SAM levels in fat cells, potentially boosting cellular energy, increasing lipid oxidation, and promoting weight loss.It is an enzyme overexpressed during skeletal muscle aging and is associated with repair of NAD+ pathway damage, dysregulated sirtuin1 activity, and increased muscle stem cell senescence. 5-Amino-1-methylquinolinium chloride promotes myoblast differentiation in vitro and enhances fusion and regeneration of muscle stem cells in aged mice. 5-Amino-1-methylquinolinium chloride is also a potent inhibitor of nicotinamide N-methyltransferase (NNMT), selectively binding to NNMT substrate binding site residues. 5-Amino-1-methylquinolinium chloride promotes myoblast differentiation in vitro and enhances fusion and regeneration of muscle stem cells in aged mice. Among them, the cytosolic enzyme nicotinamide N-methyltransferase (NNMT) has been newly discovered to regulate the levels of nicotinamide precursors required for NAD+ biosynthesis, and therefore plays a crucial role in regulating the NAD+ salvage pathway and cellular metabolism.
